Immunophilin Ligands May Prevent Antiretroviral-Associated Neuropathy

Following their development of an in vitro model of toxic neuropathy, researchers at Johns Hopkins University found that the immunophilin ligands FK506 can prevent the neurotoxicity caused by nucleoside analog reverse transcriptase inhibitors (NRTIs). In their model, Dr. Ahmet Hoke and his Baltimore, Maryland-based colleagues plated primary dorsal root ganglion neurons on a Schwann cell monolayer.

Under control conditions, most neurons bore several branching neurites after 3 hours. However, the addition of didanosine, zalcitabine or stavudine reduced the total neuritic length and the number of neurites per neuron.
