Stable paediatric renal-transplant patients can be prescribed everolimus, a novel macrolide used to prevent acute rejection in adult recipients. Paediatric patients can probably maintain the same twice-daily dosing schedule used in adult recipients, but will need a dose scaled down for body size, a phase I study by European and American researchers suggests.
Everolimus Study Group investigators from Belgium, the United Kingdom, Germany, Switzerland and four United States centres examined the single-dose pharmacokinetics, safety and tolerability of everolimus in combination with cyclosporin A and corticosteroids, both with and without azathioprine.
A total of 19 paediatric patients participated in the study. They received a single 1.2 mg/m2 dose of everolimus. The researchers report everolimus was safe and well tolerated with a low incidence of adverse events, none judged to be related to the everolimus. .
