Lamivudine is as effective a treatment as is interferon and may be better tolerated in children with chronic hepatitis.
This is the finding of an international, randomized, double-blind, placebo-controlled trial of the efficacy and tolerability of this drug for treating chronic infection with hepatitis B virus (HBV) in children, some without previous response to interferon treatment.
Maureen M. Jonas and colleagues from Children’s Hospital, Boston, United States, Diana Princess of Wales Hospital, Birmingham, England, and other centres in Europe and South and North America did this work for the International Pediatric Lamivudine Investigator Group.
In the trial, children with chronic HBV were randomly assigned in a 2:1 ratio to receive either oral lamivudine (3 mg per kilogram of body weight; maximum, 100 mg) or placeboonce daily for 52 weeks.
Of the children screened, 191 were randomly assigned to lamivudine and 97 to placebo.
The rate of virologic response was found to be 23 percent in the lamivudine group and 13
percent in the placebo group.
