Tablet Combining Glibenclamide And Metformin Improves Glycaemic Control

A tablet combining glibenclamide (glyburide) and metformin might produce better post-prandial glycaemic control compared to separate co-administration of the same agents. Researchers from Bristol-Myers Squibb Pharmaceutical Research Institute, New Jersey, United States, compared glibenclamide (2.5 mg) and metformin (500 mg) when given twice daily either together in a tablet or co-administered separately.

 The authors randomised 18 men and 19 women with type II diabetes. Thirty-five patients completed the study. Patients took the formulations for 14 days and after a two-week washout, they switched to the other formulation. Patients were aged, on average, 58 years and showed a mean body mass index of 31 kg/m2.

The glibenclamide/metformin single tablet combination resulted in a significantly smaller mean two-hour post-prandial glucose excursion (PPGE) than separate co-administration. After the standardised meal on day 14, the PPGEs were 89.5 and 117.4 mg/dl in the single tablet and separate co-administration groups respectively
