Combined cytotoxic agents temozolomide and thalidomide show antitumor activity in patients
with advanced melanoma that is well tolerated, even in those who are over age 70 years.
Researchers from the Memorial Sloan-Kettering Cancer Center, New York, United States studied 12 subjects with unresectable stage III or IV melanoma without brain metastases who were entered successively onto four treatment cohorts.
At the first level, six weeks of temozolomide 50 mg/m²/d was followed by a four week break. Temozolomide 75 mg/m²/d was for given for six weeks to levels 2, 3 and 4, followed by breaks of four, three and two weeks, respectively. Thalidomide was started at 200 mg/d and increased to a maximum dose of 400 mg/d.
