NEW YORK (Reuters Health) Apr 30 – Treatment with the monoclonal antibody trastuzumab (Herceptin) for breast cancer leads to heart failure because loss of the gene it targets, Erbb2 (Her2/neu), results in cardiomyocyte apoptosis and dysregulation of cardiac contractility.
Dr. Kuo-Fen Lee, of The Salk Institute in La Jolla, California, and associates generated mice in which the Erbb2 gene was inactivated only in the heart. At about age 6 months, these knock-out mice display ventricular enlargement and an increased ratio of heart weight to body weight compared with wild-type mice. Physiological testing of the mice showed decreased cardiac contractility.
