Tumour concentrations of urokinase-type plasminogen activator (uPA) do not predict outcome in patients with breast cancer, but concentrations of uPA inhibitors do.
S. Borstnar and colleagues from the Institute of Oncology, Ljubljana, Slovenia, measured concentrations of uPA, its receptor (uPAR) and its inhibitors (PAI-1 and PAI-2) in the tumour extracts of 460 women with primary breast cancer using enzyme-linked immunosorbent assays.
During the median 33-month follow-up period, 18.5 percent of the patients relapsed. Univariate and multivariate analyses of disease-free survival revealed that low concentrations of PAI-1 and high concentrations of PAI-2 in tumour extracts were independently linked to improved disease-free survival. When both markers were used together, their prognostic value increased further.
The only other independent predictors of disease-free survival uncovered in the study were nodal status and hormone receptor status. Concentrations of uPA and uPAR did not emerge as independent predictors of disease-free survival in the patients.
