Capsules of 9-cis-retinoic acid have moderate activity in the treatment of AIDS-related Kaposi sarcoma, say researchers in the United States. Patient responses are durable, but higher doses have major toxic effects, limiting the suitability of 9-cis-retinoic acid, report the researchers in Seattle, Washington, Tampa, Florida, Pittsburgh, Pennsylvania, and Beverly Hills and San Diego, California.
They assessed the safety, dose tolerance and anti-tumour effects of 9-cis-retinoic acid for AIDS-related Kaposi sarcoma in a phase 2, open-label clinical trial. Fifty seven men received oral doses starting at 60 mg/m² daily and rising in 40 mg. increments every two weeks up to 140 mg/m² daily.
The investigators evaluated safety by adverse events, physical examination, laboratory test abnormalities, treatment-limiting toxic effects and reasons for early withdrawal. Response, defined as 50% improvement, was gauged by overall Kaposi sarcoma response and by area and height from six index lesions at baseline….
