IL-10 often increased during migraine attack, reduced by sumatriptan treatment

WESTPORT, CT (Reuters Health) – Plasma levels of interleukin (IL)-10 were elevated in about half of subjects tested during an acute migraine attack, according to investigators in Italy. This elevation was reversed by treatment with sumatriptan.

In a study of 23 patients, 12 had plasma IL-10 levels averaging 8.83 pg/mL during migraine without aura, significantly higher than in 23 healthy control subjects (p < 0.01). Plasma levels of IL-4, IL-5, and interferon-gamma were undetectable or nearly so during the attack, Dr. Irene Munno, of the University of Bari, and associates report in the September issue of Headache.

Among 10 patients treated with sumatriptan 6-mg SQ, IL-10 levels were reduced to low or undetectable levels 30 minutes after treatment. IL-4 and IL-5 increased significantly in the same patients (p < 0.05). The latter finding is consistent with previous observations by the authors, in which plasma levels of IL-5 and IL-4 were elevated and IL-10 level was normal during the interictal period in patients with migraine.

Dr. Munno's group speculates that increased production of IL-10 during migraine may counteract the effect of proinflammatory cytokine release. Sumatriptan causes the cytokine profile to revert back to that observed in control subjects or during the interictal period.
