Interactions among insulin, insulin-like growth factor-I (IGF-I) and IGF binding proteins (IGFBPs) that increase IGF-I bioavailability may explain why environmental factors such as diet, physical activity and body size influence the risk of colorectal cancer, according to researchers.
Dr. Edward L. Giovannucci, of Harvard Medical School in Boston, and colleagues propose the novel model of colorectal carcinogenesis in a review article published in the Journal of the National Cancer Institute for July 3.
The first line of evidence is that insulin plays a role in colorectal carcinogenesis, the reviewers explain. A cross-sectional study and two prospective studies found that colorectal cancer is more common in people with hyperinsulinemia.
